Add water, stir until dissolved.<br \/>\nAdd nutrient broth.<br \/>\nHeat carefully until boiling. Don\u2019t boil over.<br \/>\nPour into Schott bottles-with the blue cap. Put cap on loosely.<br \/>\n(Or pour into conical flasks and stopper loosely with cotton wool)<br \/>\nSterilise 20mins at 151 lb\/sq in. in an Autoclave or pressure cooker.<br \/>\nCool a little and pour into sterile petri dishes. using the \u2018Aseptic Technique\u2019.<br \/>\nSeal in clear plastic bags, in the fridge. Store upside down; this stops the media from getting wet.<br \/>\nOnce inoculated, the plates must be sealed around the edges with Parafilm or wide sticky tape or completely wrapped in cling wrap.<br \/>\nInoculated plates must not be reopened.<br \/>\nDo not dispose of contaminated plates straight into the bin.<br \/>\nThe Health Department insists that THE PLATES MUST BE RE-STRERILISED IN AN AUTOCLAVE OR PRESSURE COOKER BEFORE DISPOSAL.<br \/>\nIf this can\u2019t be done then the experiment has to be cancelled.<br \/>\nPlace into an oven bag, and re-sterilise as above.
